G4X - Singular
G4X (Singular Genomics) is an in situ spatial multiomics platform that measures RNA, protein, and tissue morphology from a single FFPE section in one run, at subcellular resolution. Detection is amplification-based on padlock probes read by in situ sequencing chemistry, with a fluorescent H&E stain providing the morphology layer. A run supports up to four fluidically independent flow cells, giving up to 40 cm2 of tissue imaging and up to 128 samples, on a five-day sample-to-answer workflow. Tissue is placed onto the flow cell with the TissuStamp transfer method. The Spatial Technologies Unit operates a G4X Spatial Sequencing Center of Excellence.

Accreditations & experience
The Spatial Technologies Unit was the first site worldwide to install the G4X instrument (December 2024) and is a Center of Excellence for G4X Spatial Sequencing, designated by Singular Genomics. With three instruments, the unit can analyze more than 10,000 sections per year at 500 targets and 16 antibodies. See the Accreditations page.
Panels
G4X measures RNA, protein and morphology from the same FFPE tissue section.
- Pre-made panels: any pre-made panel, from the 200-gene base panel through any module combination, can be extended manually up to 600 targets.
- Full custom RNA: full custom panels are supported up to 500 targets.
- High-Plex custom: up to 600 targets.
- Ultra-High-Plex custom: up to 1,300 targets, available only at the STU G4X Center of Excellence.
Multi-omics
- RNA: spatial transcriptomics as described under Panels.
- Protein: up to 18-plex in situ proteomics - the 16-plex IO Protein antibody panel plus up to 2 client-selected custom antibodies.
- Direct-Seq (coming soon): in situ gap-fill sequencing of up to 100 bases of variable RNA sequence within target regions. The unit has tested Direct-Seq; it allows mutations or TCR/BCR sequences to be co-sequenced in situ.
Additional capabilities
- TissuStamp: the patented TissuStamp tissue-transfer process provides precise tissue-region selection and identical capture-region dimensions from the first to the 1,000th section.
- Flow cells per run: up to 4, fluidically independent, each with 2 or 4 lanes - up to 16 lanes and up to 128 samples per run. Each lane can run a different probe set, so different experiments run side by side without interference.
- Flow-cell configurations: the X2 flow cell supports large imaged areas - 10 x 1 cm2 (10 x 10 mm) sections, two lanes of five sections. The X4 flow cell supports high sample numbers - 32 samples of 4.5 x 4.5 mm across four lanes. Running four flow cells at once enables 40 x 1 cm2 samples (10 x 10 mm) or 128 samples of 4.5 x 4.5 mm per run.

Access and priority
Internal clients (BIDMC, DF/HCC), external academic and non-profit clients, and industry clients all have equal priority. Projects begin upon sample receipt.
